Novel aspect: A novel confirmatory method for the determination of natural hormones according to Commission Decision 2002/657/EC and the Italian legislation on natural hormones.
A recently in-house validated LC-MS/MS method for the separation, identification and quantification of residues of 17α-estradiol (17α-E2) and 17β-estradiol (17β-E2) in bovine serum is reported. To our knowledge, no citations are in literature concerning the chromatographic separation of those analytes. The method, validated according to Commission Decision 2002/657/EC, is routinely used in the Italian National Reference Laboratory for residues for the confirmatory analysis. The obtained decision limits (CCα) were of 0.06 ng/mL and 0.03 ng/mL, respectively for 17α-E2 and 17β-E2. Those values are in conformity with the Italian legislation on natural hormones. Detection capabilities (CC&beta) were obtained of 0.08 ng/mL and 0.04 ng/mL, respectively for 17α-E2 and 17β-E2. The confirmatory method was also validated through the determination of precision, accuracy, specificity, stability and calibration curve. Recovery data ranged from 86.3% to 93.2%. The method was specific and robust for the required purposes.
